I wouldn't want to write a virus, but how would one go about making an anti-virus? What to anti-virus programs check for?

An antivirus usually scans files for traces of virus code, of which it usually has several examples in its database. It may also look for changes in exe file attributes, e.g. size/date modified.
EDIT: something that would be interesting is a self-replicating antivirus...
